摘要
目的探讨5-氨基乙酰丙酸(5-ALA)诱导原卟啉IX(PpIX)在大肠肿瘤内积聚的特性。方法腹腔注射DMH溶液(1,2-二甲肼)建立大肠癌大鼠模型。尾静脉注射5-ALA溶液,对正常、早癌及进展期癌3组大肠组织分别取样,用化学抽提法提取组织原卟啉IX并定量检测其含量。结果在正常组、早癌组和进展期癌组中,大肠组织原卟啉IX含量差别均具有统计学意义,其原卟啉IX含量的平均值分别为(622.966±61.85)ng/g、(763.098±83.93) ng/g、(844.164±87.56)ng/g。结论5-ALA诱导原卟啉IX在大鼠大肠正常组织和癌组织中的积聚差异显著,为大肠早癌的荧光光谱诊断提供了实验依据。
Objective To study characteristics of different intracellular accumulation of protoporphyrin Ⅸ (PpⅨ) induced by 5-aminolaevalinic acid(5-ALA) in normal and cancer colon tissue. Methods To set up SD rat model of colon cancer by weekly intraperitoneal injections of 1,2-Syn-Dimethylhydrazine dihydrochloride (DMH) liquor. Tail intravenous injections of 5-ALA at a dose of 25mg/kg body weight after 150min,chemical extraction assays of the PpⅨ content by fluorescence spectrophotometer were carried out. Results A significant difference of PpⅨ content was found in different tissue, with a mean of (622.966 ± 61.85)ng/g, (763.098 ± 83.93) ng/g and (844. 164 ± 87.56) ng/g, respectively corresponding to normal tissue, early cancer and advanced cancer tissue in rats colon. Conclusion 5-ALA can induce effective differences of PpⅨ content in normal and cancer colonic tissue and provide experimental gist for early photie dynamic diagnosis of colon cancer.
