对苯二甲酰亮氨酸的合成研究

Research on synthesis of p-phthaloyl leucine

对苯二甲酰亮氨酸为氨基酸系表面活性剂中的一类,是由对苯二甲酰氯与亮氨酸通过非均相法合成得到.最佳反应条件为:对苯二甲酰氯与亮氨酸的物质的量比为1∶2.4,对苯二甲酰氯以2～4滴/min的速度缓慢滴入亮氨酸溶液中,反应过程中保持pH 8～9之间,反应温度控制在10～15℃.反应2h后,用浓盐酸调节pH 3～4,使钠盐转化为对苯二甲酰亮氨酸,减压抽滤得粗品.经纯化的产品收率为76.0%.

p-Phthaloyl amino acid is a sort of surface-active agent.Taking p-phthalyl alcohol and leucine as raw material, pphthaloyl leucine was obtained by nonhomogeneous reaction. The optimum conditions for the reaction were：molar ratio of the p-phthalyl alcohol and leucine was 1：2.4,the p-phthalyl alcohol was slowly added into leucine solution, the pH of the solution was kept within 8 - 9 during the reaction, the reaction temperature was controlled at 10- 15℃ .After 2 hours,the pH of the solution was adjusted to 3 - 4 by hydrochloric acid in order to transformed the sodium salt to the acid. The precipitate was collected by filtration under reduced pressure. The yield was 76 % .
