摘要
目的研究垂体瘤转化基因(PTTG)在慢性粒性细胞白血病(CML)患者中不同临床阶段的基因表达,为阐明CML演变的可能机制提供依据。方法采用半定量RT-PCR方法检测34例CML患者(慢性期16例,加速期11例,急变期7例)及10例正常骨髓单个核细胞(BMMNC)中PTTG基因的表达。结果10例正常对照中未检测到PTTG基因的表达,34例CML患者中,慢性期组、加速期组、急变期组的PTTG基因的表达分别为0.2400±0.1407,0.6509±0.2307,0.5943±0.1214,明显高于正常对照组的0.0590±0.0233(P<0.05)。PTTG基因在加速期及急变期的表达高于慢性期(P<0.05),PTTG基因在加速期和急变期的表达升高但差异无统计学意义(P>0.05)。结论PTTG基因的过度表达和CML的发生、发展有关。
Objective To explore the expression of PTTG gene of chronic myeloid leukemia(CML) in different phases for providing information about the potential mechanisms of CML. Methods Expression of PTTG gene mRNA was detected in bone marrow mononuclear cells of normal controls (BMMNC) from 34 patients with CML(16 chronic phase, 11accelerated phase and 7 blast crisis) and 10 normal controls by reverse transcriptase-polymerase chain reaction (RT-PCR). Results No detectable PTTG gene was found in BMMNC. Expression of PTTG gene mRNA in chronic phase, accelerated phase and blast crisis was 0.2400±0.1407, 0.6509±0.2307, 0.5943±0.1214, significantly higher in CML patients than in normal controis 0.0590±0.0233 (P〈0.05).The expression of PTTG gene was higher in accelerated phase and blast crisis than in chronic phase (P〈0.05), Expression of PTTG gene in accelerated phase and blast crisis showed no significance (P〉0.05). Conclusion Overexpression of oncogene PTFG gene may be related to the progression and genesis of CML.
