摘要
建立了测定样品中α吡咯烷酮乙酰胺含量的高效液相色谱法。采用的色谱柱为十八烷基硅烷键合相,硅胶为填充剂(25cm×4.6mm,粒径5μm,);流动相为甲醇乙腈水(体积比5∶2∶3);流速1mL/min;紫外检测波长230nm;内标为对乙酰氨基酚;进样量20μL;线性范围0.1~0.5mg/mL;相关系数为0.9998;回收率99.8%;相对标准偏差0.33%.与氮测定法比较,本法操作简便,结果准确。
A HPLC method for the determination of the content of 2-oxo-l-pyrrolidineacetamide in product was established, A C18 (25 cm × 4. 6 mm, 5 μm) column was used as the stationary phase, filled by silica gel; the mobile phase was composed of methanol-acetonitrile-water (5 : 2 : 3) ; at a flow rate of 1.0 mL/min and UV detection wavelength at 230 nm; paracetamol was used as internal standard; the correlation coefficient of the calibration curve was 0. 999 8 in the range of 0.1-0.5 mg/mL; the average rate of recovery was 99.8 % RSD=0. 33%. Comparing with nitrogen mensuration, the method is simple, quick and accurate.
