摘要
通过对影响RAPD标记稳定性的引物浓度梯度、退火温度梯度、dNTP浓度梯度、Taq酶浓度梯度等外部条件的优化研究,确定了RAPD反应适宜的引物浓度为0.1~0.2 pmol/μL,退火温度为48~52 ℃,dNTP浓度为0.3~0.15 mmol/L及Taq酶浓度为0.3 U/μL,提高了该反应体系中RAPD的稳定性.
Factors that affect the stability of RAPD were optimized such as primer concentration grads, annealing temperature grads ,dNTP concentration grads and taq concentration grads. The proper concentration of primer was 0.1-0.2 pmol/μL,the proper annealing temperature 48-52℃,the proper concentration of dNTP 0.3-0. 15 mmol/L,the proper Taq concentration 0. 3 U/μL. and the stability of RAPD was enhanced in this system.
