马钱子对三种软骨细胞凋亡模型的影响

Effects of Semen strychni on three types of chondrocytes apoptosis

目的:观察中药马钱子对三种软骨细胞凋亡模型的影响。方法:在兔软骨细胞培养体系中分别加入2mmol/L硝普钠(SNP)、2mmol/LSNP+马钱子、10-4mol/L全反式维甲酸(ATRA)、10-4mol/LATRA+马钱子;在人胚胎软骨细胞培养体系中加入30ng/L肿瘤坏死因子α(TNFα)、TNFα+马钱子,各组培养24h后,采用AnnexinV/PI双参数法对软骨细胞凋亡进行定量检测。结果:软骨细胞在加入SNP、ATRA、TNFα发生凋亡时,加入马钱子能降低SNP、ATRA诱导的软骨细胞凋亡率(P<0.01,P<0.05),但对TNFα诱导的软骨细胞凋亡,无明显抑制作用(P>0.05)。结论:中药马钱子对部分软骨细胞凋亡有一定抑制作用。

Objective.To evalute the effects of Semen strychni on three types of chondrocytes apoptosis. Methods： Chondrocytes of rabbits were respectively cultured with ①2 mmol/L sodium nitroprusside（SNP）, ② Semen strychni and 2 mmol/L SNP, ③10^-4 mol/L all-trans retinoic acid （ATRA）, ④ Semen strychni and 10^-4 mol/L ATRA. Fetac chondrocytes were respectively cultured with ①30 ng/L tumor necrosis factor-α （TNF-α）, ②Semen strychni and 30 ng/L TNF-α. The rates of chondrocytes apoptosis in every group were detected by Annexin V/PI with flow cytometry after being cultured for 24 h. Results：The chondrocyte apoptosis could be induced by SNP,ATRA and TNF-α respectively. Adding Semen strychni into cultured could decrease chondrocytes apoptosis induced by SNP,ATRA（P 〈 0.01, P 〈 0.05）, but had no effects on apoptosis induced by TNF-α（ P 〉 0.05 ）. Conclusion： Semen strvchni has inhibitorv effects on some chondrocyte apoptosis.
