小麦蓝矮病植原体的分子生物学检测研究 被引量:1

摘要 小麦蓝矮病是中国北方小麦的重要病害。利用植原体特异引物对小麦蓝矮病带病植株总DNA进行巢式-PCR(Nested-PCR)扩增,获得1.2kb的特异性片段,从分子水平上证明小麦蓝矮病是一种植原体病害。对具有红矮和蓝矮症状的病株扩增均可扩增出目标片段,说明红矮和蓝矮症状都是由植原体引起。试验为在小麦蓝矮病分子诊断、田间介体带毒检测、寄主范围研究等提供一种快速、灵敏可靠的方法。 Wheat Blue Dwarf is one of the main disease on winter wheat in Northwestern China. A 1.2kb fragment was amplified by phytoplasmas universal primers in total DNA sample extracted from diseased wheat with dark green leaves and red dwarf symptom by Nested-PCR. The result indicated molecularly that the pathogen of Wheat Blue Dwarf disease was phytoplasma. The different symptoms (dark green dwarf and red dwarf) were all caused by phytoplasmas. Nested-PCR could be as a rapid, exact technique for detecting the pathogens from wheat leaves, insect vectors and weed host.
