nm23-H_1基因在急性白血病中表达及与多药耐药关系 被引量:2

EXPRESSION OF nm23-H_1 IN ACUTE LEUKEMIA PATIENTS AND ITS CORRELATION WITH MULTIDRUG RESISTANCE

摘要 ①目的 检测急性白血病病人nm23- H1 基因表达,探讨其与病人预后和多药耐药的关系。②方法采用半定量逆转录聚合酶链反应(RT PCR)技术,检测46 例初诊急性白血病病人及15 例正常对照组骨髓细胞nm23 H1 表达,同时检测其多药耐药基因MDR1及MRP表达。③结果 正常骨髓细胞nm23 H1 表达阴性;46 例急性白血病病人中,43例有nm23- H1 表达,且其表达水平与MDR1 及MRP 表达水平无明显相关性(r= 0.39、0.21,P>0.05)。④结论 nm23 -H1 高表达与急性白血病发生有关,但与MDR1及MRP基因的表达无关。 Objective To determine the expression of nm23-H1 gene in acute leukemia patients and to evaluate the relationship between nm23-H1 expression and multi-drug resistance. Methods Expressions of nm23-H1 mRNA, MDR1 and MRP were determined in bone marrow cells from 46 acute leukemia patients and 15 normal subjects by semi-quantitative reverse transcription polymerase chain reaction(RT PCR). Results nm23-H1 was negative in 15 normal subjects, while it was positively expressed in 43 of 46 acute leukemia patients. The level of nm23-H1 expression had no significant correlation with that of MDR1 or MRP(r=0.39,0.21,P〉0.05). Conclusion The high nm23-H1 gene expression is related to the occurrence of acute leukemia, but has no correlation with MDR1 or MRP.
