黄芪三仙汤通过海马—NPY—PAA对调节细胞免疫功能的观察

摘要 目的观察黄芪三仙汤通过海马—NPY—PAA对调节细胞免疫功能的影响。方法将24只雄性大鼠随机分为3组,即假手术组、模型对照组和治疗组,各组8只。将1μlβ-淀粉样蛋白1-40(Aβ1-40)(10μg/μl)在立体定向仪下注入大鼠右侧Meynert核(NBM)制成阿尔茨海默病(AD)模型,治疗组用黄芪三仙汤灌胃,模型对照组和假手术组给予0.9%氯化钠注射液灌胃,4周后观察海马区神经肽Y(NPY)含量,血浆促肾上腺皮质激素(ACTH)、血清皮质酮(CORT)含量、胸腺指数、脾脏巨噬细胞数目的变化。结果假手术组、治疗组海马NPY含量,血浆ACTH,血清CORT,脾脏巨噬细胞数较模型对照组显著升高(P<0.01),胸腺指数较模型对照组无显著变化(P>0.05)。结论黄芪三仙汤可以通过海马—NPY—垂体—肾上腺轴调节细胞免疫功能。黄芪三仙汤提高Aβ1-40注射模型大鼠的认知功能可能与其增加NPY的释放有关。 Objeaive To investigate the neural immune modulation mechanism affected by Huangqisanxian decoction on treating Alzheimer's disease(AD). Methods 24 male rats were randomly divided into three groups: the pseudo-operation, the model control and the treatment group. 8 rats were in each group . Amyloid-beta protein (10 μg) were injected into the right Meynert nucleus modulated by the stereotacic apparatus to make the AD model rats. The rats in the treatment group were orally administrated huangqisanxian decoction. The rats in the pseudo-operation group and model contral group were given 0.9% Nacl injection. The mount of NPY, plasma ACTH level, serum cortisone level, thoracic gland index and the number of spleen macrophage were observed after four weeks. Results The mount of NPY, plasma ACTH level, serum cortisone level and the number of spleen macrophage in the pseudo - operation group and treatment group were significantly higher than those in the model contral group ( P 〈 0.01 ). There was no significant difference of thoracic gland index in the three groups. Conclusion Huangqisanxian decoction can adjust immune function by affecting hippocampus - NPY- PAA and can improve the cognition of rats in the model group. The potential mechanism was the increasing level of NPY.
