高效液相色谱法测定连胶素胶囊中连翘苷的含量

Determination of Forsythin in the Lianjiaosu Capsule with RP-HPLC

目的建立连胶素胶囊中连翘苷含量的HPLC法。方法反相高效液相色谱法。采用HypersilODS C18色谱柱;以乙腈-水(30:70)为流动相;流速为1.0ml/min;柱温为室温;检测波长为277nm;进样体积10μl;外标法计算含量。结果连翘苷在0.1875μg-1.25μg范围内峰面积与进样量呈良好线性关系,其回归方程为A=715534.72C-7249.55,r=0.9998,其平均回收率为99.27%,RSD为3.22%(n=5),测得连胶素胶囊中连翘苷的含量为0.2089mg/g。结论本方法简便、准确,提供了连胶素胶囊中连翘苷的质量控制方法。

Objective To establish a quantitative method to determine the content of Forsythin in the Lianjiaosu capsule. Methods The content of Forsythin in the Liaojiaosu capsule was determined wioth RP- HPLC with Hypersil C18 column at room temperature, The mobile phase was aeetonitrile-water（30： 70）with a flow rate of 1.0ml/min and the injection volume was 10uL The external standard was used and the wavelength of the detector was set at 277nm, Results The linear ranger was 0. 1875μg-1.25μg and correlation efficient was 0.9998. The average recovery and the relative standard deviation of Forsythin in Lianjiaosu capsule were 99.27% and 3.22%. Conclusion The method is convenient ,accurate, and can be used for the de termination of Forsythin in Lianjiaosu capsule.
