摘要
本文在实验的基础上对硫酸-苯酚定糖法进行了改进。传统的硫酸-苯酚定糖法,平行样品间的吸光度值相差较大,使检测的重现性与准确性较差。我们对该方法进行改进后,并对新方法从检测波长、显色时间、显色温度、稳定性、重现性、回收率、线性范围以及最低检出限等方面进行了考查,并通过对纯化的银耳多糖、可溶性淀粉、猪苓多糖和海带多糖,以及对猪苓与海带水溶性混合物等多种样品的多糖含量测定,证明改进后的方法既简化了操作,提高了精密度和准确性;也具较宽的实用范围。
The article improved Phenol-Sulphate acid method on the base of experiment. If we adopt traditional Phenol-Sulphate acid method, the deference of parallel sample's absorbency is larger and the accurate and reproducible is worse. We used the new method to checked inspected wavelength, color test time, color test temperature, stability, reproducible, recovery ration, linearity scope and the limit of determination. Also we examined polyaccharide of purified Tremella polysaccharide, soluble starch, Polyporus umbellatus ploysaccharide, and laminarin, and other polysaccharide samples. All what we have done prove that the improved method can simplify the operation, increase accurate and reproducible.
