白细胞介素-8及其受体在小儿急性白血病骨髓单个核细胞中的表达和临床意义

摘要 目的探讨白细胞介素8(IL8)及受体(IL8R)在小儿急性白血病骨髓单个核细胞中的表达和临床意义。方法RTPCR法检测32例急性白血病骨髓单个核细胞IL8,IL8RmRNA表达。结果急性髓系白血病(AML)组中有5例表达IL8(5/10),4例表达IL8R(4/10);急性淋巴细胞白血病(ALL)组中有13例表达IL8(13/22),9例表达IL8R(9/22)。IL8在急性髓系白血病组中的表达水平高于急性淋巴细胞白血病组,差异有显著性(P<0.05)。AML组中M4M5亚型中的表达水平高于M2M3,差异有显著性(P<0.05)。ALL组中B系列ALL表达水平高于TALL,差异有显著性(P<0.05)。化疗后患儿骨髓单个核细胞IL8,IL8R水平显著低于化疗前(P<0.05)。结论小儿急性白血病有IL8,IL8R的表达,IL8尤其在AML组M4M5亚型及B系列ALL更明显,化疗后两者水平显著下降。 Objective To study the expression levels of interleukin-8 ( IL-8 ) and its receptor in bone marrow cells of children with acute leukemia. Methods The expressions of IL-8 and its receptor in bone marrow cells were detected using RT-PCR in 22 cases of acute lymphoblastic leukemia (ALL) and 10 cases of acute myelocytic leukemia (AML). Results In 5 of 10 AML patients and in 13 of 22 ALL patients, the cells expressed IL-8 mRNA. In 4 AML cases and in 9 ALL cases, the cells expressed IL-8 receptor mRNA. The IL-8 expression levels in AML patients were significantly higher than those in ALL patients( P 〈 0.05 ). The cells from patients with M4-M5 AML expressed a higher IL-8 mRNA level than those from M2-M3 AML patients ( P 〈 0. 05 ). Of the ALL children, the IL-8 expression levels in B-ALL children were significantly higher than those of T-ALL children(P 〈 0.05 ). After induction chemotherapy, the expression levels of IL-8 and its receptor in bone marrow cells of children with acute leukemia were significantly reduced( P 〈 0.05 ). Conclusions IL-8 and its receptor may be expressed in childhood acute leukemia. Children with M4-M5 AML or B-ALL can express a higher level of IL-8 mRNA in bone marrow cells. Induction chemotherapy can deerease the expression of IL-8 and its receptor.
