用荧光钙离子指示剂fura-2AM检测心肌细胞内游离钙

Test of the intracellular free calcium in rat insolated ventricular myocytes using fura-2AM fluorescence free calcium indicator

本实验采用游离钙离子荧光指示剂fura－2AM检测大鼠离体心室肌细胞内游离钙的浓度。其结果如下；（1）KC1可以使心肌细胞内游离钙浓度升高，去掉细胞外液中钙离子，KC1的升钙作用则消失。（2）甲氧胺和咖啡因可使心肌细胞内游离钙浓度升高。二者的作用分别被哌唑嗪和普鲁卡因所对抗。提示该法切实可行。

A method to test the intracellular free calcium (Ca^(2+))i in insolated rat ventricular myocytes using fura-2AM, a intracellular free calcium indicator, was recommeded.Using this method, it was showed that KCL (40mmol/L), Methexamine(40mmol/L) and Caffeine (5mmol/L) increased the (Ca^(2+))i in insolated rat ventricular myocytes, their effects were abolished by extracellular Ca^(2+)-depriration additions of prazosin and procaine, expectively. The results suggest that the method is reliale in testing (Ca^(2+))i.
