机械应力刺激对体外肺腺癌细胞增殖的影响

Effect of mechanical strain on proliferation in lung adenocarcinoma cells in vitro

摘要 通过FX-4000柔性基底加载系统研究了不同波形的周期性拉伸对人肺腺癌A549细胞株增殖的影响,应用Image-pro图像处理软件对A549细胞株在心形波、三角波及方波等拉伸应变下的增殖动力学变化进行了分析.实验结果表明:在collagen Ⅰ基底膜上,应变为0%~20%,频率为0.5Hz及1.0Hz,加载时间为2h时,与对照组比较,方波刺激组细胞生长明显受到抑制,三角波与心形波刺激组细胞增殖率无明显差异.研究表明:A549细胞株对体外的生理应变作出响应时,方波的抑制作用最佳,而拉伸频率的影响并不明显. Proliferative responses of human lung adenocarcinoma cell line A549 to cyclic mechanical strain with varying waveforms were studied by FX-4000 strain unit. The results showed that cellular proliferation rate(PR) reduced significantly in the squarewave stretch group compared with the control after A549 cells were subjected to 0 - 20% elongation at frequency 0.5Hz or 1.0Hz for 2h. The PR had no significant difference in the trianglewave and heartwave stretch groups although the cells were stretched for 2h. It is concluded that A549 cells do respond to physiological strain in vitro, squarewave stimulation significantly repressed A549 cells proliferation, effects of strain frequency had no significant difference between 0.5Hz and 1.0Hz. The selected waveform play an important role for repressed A549 cells proliferation.
