摘要
在工程菌株JM109中实现了带有6×His标记的新型重组别藻蓝蛋白HAPC的高效表达.工程菌的发酵采用两步法,37℃扩增生长6h,30℃诱导培养10h.发酵液最终菌体密度(OD600)达26.25,表达蛋白占菌体总蛋白的31%.进一步用金属螯合亲和层析纯化使其纯度达90%以上.用纯化的HAPC对S180荷瘤小鼠灌胃,每天1.5~4.5mg/kg,共10天.结果表明HAPC具有明显的升白作用,可对抗环磷酰胺对免疫系统的破坏作用.
A new 6 × His· tag recombinant allophycocynin HAPC was efficiently expressed in E. coli strain JM109. During the fermentation, the strain grew in 37℃ and then was induced in 30℃. After 16h fermentation, the final cell density reached OD600 26.25 with expressed HAPC to 31% of the total. The protein was purified to the purity of over 90% by metal chelate chromatograph. Stso tumor carried mice were treated with purified HAPC ig 1.5 - 4.5 mg/kg per day for 10 days. The result showed that HAPC, with obvious leukogenic effect, could inhibit immune damage of antitumor chemical cyclophosphamide (CTX).
