摘要
目的:探讨水蛭注射液对缺血再灌注大鼠脑的保护机制。方法:采用颈总动脉栓线法造成大鼠脑缺血再灌注模型,观察缺血脑组织内白细胞浸润情况及脑梗塞面积的变化,检测大鼠脑匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)、血清肿瘤坏死因子(TNF)的变化。结果:缺血24h时药物组脑匀浆SOD的活性显著高于对照组(P<0.01),而药物组的脑匀浆MDA和血清TNF的浓度明显低于对照组(P<0.05),药物组的脑梗塞区内白细胞浸润及脑梗塞面积也明显小于对照组(P<0.01)。结论:水蛭注射液对缺血再灌注大鼠脑有保护作用,其机制可能与抑制白细胞的浸润、减少自由基的生成有关。
Objective: To study the mechanism of protective function of hirudo injection on ischemia - reperfusion injury of brain in rats. Methods: The model of ischemia - reperfusion injury of brain in rats was made by inhibiting the common carotid artery with thread, leukocyte infiltration in ischemia cerebral tissues and the change of cerebral infarction area were observed, and the alteration of rats'cerebral homogenesis plasma ultra - oxides, SOD, MDA and TNF, were determined. Results: After 24h ischemia, cerebral homogenesis plasma SOD activity in the hirudo treatment group was remarkably higher than that of the control group(P 〈 0.01), while the concentration of cerebral homogenesis plasma MDA and serum TNF in the hirudo treatment group sharply dropped (P 〈 0.05). Compared with the control group, the number of leukocyte infiltration and the infarcted area in the hirudo treatment group greatly dropped too ( P 〈 0.01). Conclusion: Hirudo injection maybe has the protection against ischemia - reperfusion injury of brain in rats by inhibiting leukocyte infiltration and reducing the production of free radical.
