小剂量骨髓细胞联合脾细胞输注调节受体特异性免疫状态的研究

摘要 目的建立供体小剂量骨髓细胞输注同时联合脾细胞输注对受体特异性免疫状态的调节方法,建立相应的评估体系,探索其调节效应。方法C57BL/6小鼠输注小剂量BALB/C小鼠骨髓细胞或骨髓细胞脾细胞联合输注,观察受体存活状况,3个月后建立混合淋巴细胞培养体系,评估受体特异性免疫状态。结果受体存活好,移植物抗宿主反应强度较低,混合淋巴细胞反应联合输注较骨髓细胞输注稍低但差异无显著性。结论供体骨髓细胞联合脾细胞输注可以在减少骨髓细胞输注量时仍有效调节受体特异性免疫状态,混合淋巴细胞培养体系是一种有价值的评估方法。关键词:骨髓细胞输注;脾细胞; [Objectives] To establish the donor splenocyte infusion combining bone marrow infusion to modulate the specific immune response in recipients and the system to evaluate. [Methods] C57BL/6 mice received the lowdose BALB/C mice bone marrow or combining splenocyte. The survival status was observed.3 months post infusion the mixed lymphocyte reaction was established to evaluate the donor-specific immune reaction. [Results] The recipients survived well and there was no graft-versus-host-disease. Mixed lymphocyte reaction was lower in the combining infusion than bone morrow single infusion but no significant difference was observed. [Conclusions] The donor splenocyte infusion combining lower dose bone marrow infusion can modulate the specific immune response in recipients, and the mixed lymphocyte reaction is a valuable system to evaluate the effect and the immune status.
