基质金属蛋白酶-9和原癌基因c-myc在甲状腺癌中的表达及其关系探讨 被引量:4

摘要 目的探讨基质金属蛋白酶-9(MMP-9)和原癌基因c-myc在甲状腺癌中的表达及其关系。方法手术中获得的甲状腺癌、甲状腺腺瘤、桥本氏甲状腺炎及正常甲状腺组织的新鲜标本共39例,提取其总RNA,然后进行半定量逆转录多聚酶链反应(RT-PCR),比较MMP-9及c-mycmRNA在不同疾病组织中的表达差异。用酶联免疫吸附方法(ELISA)检测不同患者血清中的MMP-9水平。结果在甲状腺癌组织中,MMP-9及c-mycmRNA的表达均显著性高于甲状腺腺瘤、桥本氏甲状腺炎及正常甲状腺组织(P<0.05),其他3组间的比较无统计学意义。甲状腺癌患者同时伴有血清MMP-9水平的显著性升高。结论MMP-9及c-mycmRNA的表达增高及血清MMP-9水平的升高可能与甲状腺癌的发生有关。 [Objective] To investigate the expression of matrix metalloproteinase 9 (MMP-9) and c-myc genes in thyroid carcinoma and their relation in the pathogenesis of thyroid carcinoma. [Methods] The thyroid tissues were selected in surgical operation, including 9 thyroid carcinoma cases, 8 Hashimoto's thyroiditis cases, 11 thyroid adenoma cases and 11 normal tissues. The total RNA was extracted and gene expression of MMP-9 and c-myc was evaluated by RT-PCR. The serum MMP-9 levels were determined with ELISA assays. [Results] The levels of MMP- 9 and c-myc mRNA in thyroid carcinoma group were significantly higher than those in other 3 groups (P 〈0.05). Also, the serum MMP-9 levels in patients with thyroid carcinoma were significantly increased. [Conclusions] The high expression of MMP-9 and c-myc mRNA and serum MMP-9 levels appear to be related with the pathogenesis of thyroid carcinoma.
