丙泊酚、咪唑安定对心肌缺血再灌注损伤Fos蛋白表达的影响 被引量:1

摘要 目的:观察丙泊酚、咪唑安定在心肌缺血再灌注损伤(IR)时Fos蛋白表达的影响,从蛋白表达水平探讨其对心肌保护作用的机制。方法:健康SD大鼠36只,每组12只。分别给生理盐水(A组)、丙泊酚(B组)、咪唑安定(C组)静脉输注,在心肌缺血15min后,恢复心肌血液灌注,分再灌注0、15、30、60、120、240min6个时相,各时相取2只大鼠,多聚甲醛灌注固定后取缺血心肌标本作石腊切片,免疫组化方法检测Fos蛋白表达,显微镜下观察阳性产物,拍片。结果:各组组内再灌注30、60、120min阳性表达灰度值较再灌注0、240minFos蛋白表达明显增强(P<0.05);A组再灌注30、60、120minFos蛋白表达均比B、C组增强(P<0.05),统计学有显著性意义。结论:丙泊酚、咪唑安定对心肌缺血再灌注损伤有一定的保护作用,能降低缺血再灌注心肌Fos蛋白表达,为心肌缺血再灌注损伤时安全可用的麻醉药,其作用与缺血再灌注时间有关,即在30~120min内对缺血再灌注心肌有明显保护作用,这对临床选择合理时间窗用药有一定的指导意义。 Objective:To observe the effects of propofol and midazolam on the expression of Fos protein during myocardial ischemia and reperfusion injury,try to find propofol,midazolam the relationship of protective myocardial ischemia in protein lever. Methods: Thirty-six adult SD rats,the left anterior coronary descending branch were ligated in 15 min,then the ligation were removed,and divided into 3 groups: group A eontrol(NaCl),group B (propofol), and group C (midazolam). All groups divided 6 time phase got myocardial tissue IR 0, 15,30,60,120, and 240 min. Immunohistochemistry technique was used to analyze the amount change of the Fos protein express. Results: Fos protein expression gray change,each group IR 30,60,120 min Fos protein gray change was higher than IR 0,240 min(P 〈 0.05):Group A Fos protein express were significant highter than group B and group C in IR 30,60, and 120 min(P 〈 0.05). Conclusion: Propofol and midazolam decease myocardical ischemia reperfusion injury Fos protein expression and protect myocardical function This protect function was corresponded with reperfusion time, and these were important for choocing exact time phase to use drugs in clinic work.
