高羊茅成熟种子组织培养的影响因素研究 I.多种因素对愈伤组织诱导的影响 被引量:6

摘要 以4个草坪型高羊茅品种的成熟种子为外植体,研究多种因素对愈伤组织诱导的影响。结果表明,增加2,4-D浓度能显著提高高羊茅成熟种子愈伤组织诱导率,而同时辅之以ABA则能获得更好的诱导效果,其最佳组合为8mg/L2,4-D+2mg/LABA;种子灭菌后纵切或切胚可使出愈率成倍提高;采用MS基本培养基和在培养基中添加水解酪蛋白与谷氨酰胺也有助于提高出愈率;诱导培养基中添加BAP在多数情况下导致出愈率降低,但对改善愈伤组织质量有利。 The influences of multiple factors on callus induction from mature seeds of four tuff-type tall fescue cultivars were studied. The results showed that enhancement of 2,4-D concentration in medium significantly increased callus induction rate. Combination of 2,4-D and ABA could produce better induction effects, with optimum combination being 8 mg/L 2,4-D + 2 mg/L ABA. Longitudinal slicing or embryo cutting of sterilized seeds resulted in one and half to eight times higher callus induction rate over intact seeds. Adoption of MS basal medium and addition of casamino acids and glutamine 0.5 g/L each also helped facilitate callus formation. Addition of BAP in the induction medium decreased callus induction rate in most cases, but was beneficial to improvement of quality of induced calli.
