以MTT比色法检测鸡脾淋巴细胞转化效果

MEASUREMENT OF CHICKEN SPLENIC LYMPHOCYTE ACTIVATION BY MTT COLORIMETRIC ASSAY

本研究运用ＭＴＴ比色法以正交试验对鸡脾脏淋巴细胞转化的最佳条件（ｃｏｎＡ量、血清种类和浓度）进行了研究。结果表明，无论血清种类和浓度如何，低剂量ＣｏｎＡ（２．５～１０μｇ／ｍｌ）可以获得较好的转化效果。当ＣｏｎＡ的量确定时，低浓度鸡血清（０．５％～１．２５％）的转化效果优于高浓度（２％～３％）：５％犊牛血清优于任何浓度的鸡血清；无血清培养基优于血清培养基。其最佳组合为无血清ＲＰＭＩ１６４０培养基加ＣｏｎＡ２．５μｇ／ｍｌ。

MTT colorimetric assay was used to measure the perfect conditions(ConA concentrations,kinds andconcentrations of serums)for chicken splenic lymphocyte activation in orthogonal experiment. It was found that lowdoes ConA(2.5～10μg/ml)with 0～3%chicken serum or 5%fetal calf serum could resulted in a good activation incontrast with high dose ones(20～60μg/ml ).Whcn the conccntration of ConA was determined,Iow concentrationchicken serum(0～1.25%)induced higher activation than that induced by high ones(2～3%).The activationrcsulted by fetal ealf serum,howerver,was better than that resulted by chicken scrum of any concentrations. Themore perfect conditions for activation were RPMI 1640 medium with 2.5μg/ml ConA.
