唐山市流行性腮腺炎病毒小疏水蛋白基因序列分析 被引量:3

摘要 目的探讨唐山市流行性腮腺炎病毒的分子特征.方法收集唐山市不同行政区的流行性腮腺炎患儿唾液标本5份,其中,爆发病例2例,散发病例3例.经RT-PCR扩增小疏水蛋白(SH)基因并测序,利用Clustal X软件进行系统发育分析.结果唐山市分离的5株流行性腮腺炎病毒均为F亚型.各株病毒SH基因的核苷酸序列一致率为94.8%~100.0%,氨基酸序列一致率为91.4%~100.0%,但与国内主要野毒株SH基因的氨基酸序列一致率只有80.7%.系统发育分析显示,唐山市的各样本之间同源性较高,其次是国内的野毒株,而与国外野毒株及疫苗株同源性低.结论唐山市存在多种流行性腮腺炎病毒株流行,主要基因型为F亚型. Objective To explore the molecular chracteristics of mumps virus in Tangshan city. Methods 5 saliva specimens were collected from 5 clinical mumps cases in different districts of the city. Two of them came from two outbreaks respectively, others were sporadic. Mumps virus RNA was extracted from saliva specimens by Trizol and Chloroform. A reverse transcriptase nested polymerase chain reaction was developed to detect the small hydrophohic(SH) protein gene of mumps virus. Phylogenetic analysis was performed on the entire SH gene sequence with Clustal x software. Results All of the five mumps virus strains were F subtype in the city. The five strains of genotype F were closely related to each other by 94.8 - 100% nucleotide similarity, 91.4 - 100% amino acid similarity. Compared with other wild - type strains and vaccinal strains of different areas in China, nucleotide similarity rates were 90.8 - 98.3% and 87.4 - 92.5% respectively. The phylogenetic trees based on 316 nt of the SH gene indicated that 5 strains from Tangshan group into F genotype, showed closely similarity. But the similarities were slightly lower between wild- type strains of different areas, and it was the lowest in Tangshan strains, vaccinal strains and foreign wild- type strains. Conclusion There are a lot different mumps virus strains in Tangshan city, and subgroup F is the major genotype.
