摘要
目的观察雪旺细胞(SCs)在体外对纹状体生长的影响。方法将SCs分别与胎鼠、新生鼠及成年鼠的纹状体在体外进行共培养,分别在3d,1、2、3、4周进行观察纹状体周围神经突起的生长及细胞的迁移情况。结果共培养组中,胎鼠纹状体周围在第3天就出现了神经突起的生长,第3周达到高峰;新生鼠纹状体周围在第1周出现了神经突起;成年鼠纹状体在第2周出现神经突起的生长。共培养组中,胎鼠纹状体周围第1周出现细胞的迁移,随后逐步增多;新生鼠纹状体周围第2周出现细胞的迁移;成年鼠纹状体在第3周出现少量的细胞迁移,但与对照组比较差异亦有统计学意义(P<0.05)。结论SCs在体外能促进共培养不同发育阶段纹状体神经突起的生长及细胞的迁移,可用于神经退行性疾病的移植治疗。
Objective To explore the effect of Schwann cells (SCs) on the striaturns growth in vitro. Methods SCs were co-cultured with striaturns coming from embryonic, new and grow-up SD rats in vitro. The growth of neurites around the striaturns and the migration of cells from the striaturns were observed at 3rd day, and 1st, 2nd, 3rd, 4th week. Results In co-culture group, the neurites around the the striaturns of embryonic, new, grow-up rats appeared at 3rd day, and 1st, 2nd week respectively. The migration of the cells from the striatums of embryonic, new, grow-up rats appeared at 1st, 2nd, 3rd week respectively. The number of cells moving from the striaturns of the grow-up rats were small, but it is still obvious compared to the control group (P〈 0.05). Conclusion SCs have the ability to promote the neurites growth of striaturns and cells moving from striaturns, and they can be used for cell transplantation therapy to neurodegenerative diseases.
