摘要
建立了新生小鼠颅盖骨器官培养方法,采用原子吸收分光光度技术测量培养上清液的钙含量,计算实验组/对照组的比值作为骨吸收指数。同时利用放射免疫分析方法测定上清液骨钙素含量,结合形态学观察,综合评价了培养颅盖骨的吸收活性,使颅盖骨吸收活性的检测更为准确可靠。
Weestablishedtheinvitroculturesystemofneonatalmousecalvaria.Boneresorp-tionwasdeterminedbythefolowingexaminations:(1)thereleaseofcalciumfromthecalvariaintothemediumwasmeasuredbyuseofatomicabsorptionspectrophotometer,andthetreatedtocontrolratiowascalculatedandusedasanindexofboneresorption;(2)theconcentrationofosteocalcinintheculturedmediumwasdeterminedbyradioimmunoasay;(3)histologicalexamination.Bycom-bineduseoftheexaminations,wewereabletoobjectivelydeterminetheboneresorptionofculturedcalvaria.
