摘要
高剂量大容积5-Fu腹腔给药后240分钟内腹腔液浓度是股静脉血浓度的288倍,门静脉血浓度是股静脉血浓度的13.8倍,肝静脉血浓度是股静脉血浓度的3.7倍,组织中肝浓度最高,胃、结肠次之,肺、肾最低。经腹腔化疗后的腹腔荷人结肠癌移植瘤裸鼠对照组全部产生腹腔移植瘤,5-Fu10和20mg/kg组均有2/5产生腹腔移植瘤,30mg/kg组无腹腔移植瘤产生。结果表明高剂量大容积5-Fu腹腔化疗有利于胃肠恶性肿瘤术后腹腔复发和肝转移的防治。
After IP administration of a bolus of large dose of 5-Fu, the drug concentration in peritoneal fluid,portal vein, hepatic vein were 288,13. 8,3. 7 times that of the femoral vein within 240 minutes respectively.The concentration of 5-Fu was highest in the liver, followed by the stomach,colon,lungs and kidneys in that order. A daily IP administration of 5-Fu was started on the following day after inoculating 5×10 ̄5 HCC cells into the peritoneal cavity of nude mice for 5 days.The result showed that all the control animals developed disseminated peritoneal growth of tumor, 2 of the nude mice in both 5-Fu 10mg/kg and 20mg/kg groups developed peritoneal growths, but no growth developed in the 5-Fu 30mg/kg group.That means that IP administration of 5-Fu is of advantage in preventing and treating tumor recurrence in the peritoneal cavity and hepatic metastasis after gastrointestinal tumor surgery.
