摘要
目的探讨雄激素脱氢表雄酮(DHEA)对氧化低密度脂蛋白(ox LDL)诱导的人脐静脉内皮细胞(HUVECs)表达血管细胞黏附分子1(VCAM1)的影响。方法体外培养HUVECs,采用免疫细胞化学、Western blot、原位杂交等方法,观察不同浓度DHEA(1,5,50μmol/L)对ox LDL诱导的HUVECs表达VCAM1的影响。结果ox LDL诱导培养HUVECs后,HUVECsVCAM1表达明显升高,预先用DHEA处理HUVECs可使VCAM1的表达降低(P<0.01),且这种降低呈浓度依赖性。结论DHEA能浓度依赖性地抑制ox LDL诱导的HUVECsVCAM1的表达。
Objective To investigate the effects of dehydroepiandrosterone (DHEA) on ox-LDL-induced VCAM-1 expression in human umbilical venous endothelial cells (HUVECs) . Methods HUVECs were cultured in vitro and the effects of DHEA (1, 5, 50μmol/L) on ox-LDL-induced VCAM-1 expression in HUVECs were observed by immunocytochemistry, Western blot, in situ hybridization. Results Ox-LDL stimulation significantly induced the VCAM-1 expression in HUVECs. When HUVECs were pretreated with DHEA, ox-LDL-induced VCAM-1 expression was obviously decreased in a dose-dependent manner (P〈0.01). Conclusion DHEA could inhibit the ox-LDL-induced VCAM-1 expression in HUVECs in a dose-dependent manner.
