摘要
目的探讨牛膝多糖对外周血单核细胞是否具有激活作用。方法分别应用中性红试验检测牛膝多糖(1.250mg/ml)对单核细胞吞噬作用的影响;非特异性脂酶染色及电镜技术检测牛膝多糖(1.250mg/ml)诱导单核细胞内溶酶体的改变;0.312、1.250、5.000mg/ml牛膝多糖对体外培养的外周血单核细胞进行诱导培养12h,ELISA法测定单核细胞肿瘤坏死因子α(TNFα)和白细胞介素6(IL6)的表达。结果牛膝多糖能增强单核细胞的吞噬作用,增加单核细胞胞质内溶酶体量,显著诱导单核细胞表达TNFα和IL6。结论牛膝多糖对单核细胞具有激活作用。
Objective To study the activated effects of achyranthes bidentata polysaccharides (ABPS) on human monocytes. Methods The effects of ABPS (1.250 mg/ml) on phagocytosis, lysosome of human monocytes were studied by neutral red test, and no-specific lipase staining and electron microscopy respectively. After the monocytes were cultured with 0.312, 1.250, 5.000 mg/ml ABPS-RPMI1640 for 12h in vitro, the expression levels of tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) in the monocytes were detected by ELISA. Results The phagocytosis and number of lysosomes were increased in the monocytes induced by ABPS, and ABPS could significantly induce the expression of TNF-α and IL-6 in the monocvtes. Conclusion ABPS can activate the monocvtes.
