归芪口服液促进放射损伤小鼠骨髓造血机能的恢复及其机制

摘要 目的探讨归芪口服液对放射损伤小鼠骨髓组织中CD54表达的影响及其促进早期骨髓造血恢复的可能机制。方法将42只清洁级近交系健康昆明小鼠随机分为3组。正常组不做任何处理;归芪组和对照组于6.0G60Coγ射线一次性全身均匀照射后,分别被胃饲归芪口服液(200mg/次,2次/d)和相同剂量的生理盐水。照射后第4、8和14天计数小鼠骨髓单个核细胞和骨髓组织中巨核细胞数,测量骨髓造血组织面积,并用免疫组织化学法检测骨髓组织中黏附分子CD54的表达水平。照射后第8天计数小鼠的脾集落形成单位(CFU S)数。结果照射后第48、14天归芪组骨髓单个核细胞、骨髓组织中巨核细胞计数和骨髓造血组织面积均显著高于对照组(P<0.01或P<0.05);第4、8天归芪组骨髓组织中CD54的表达水平显著高于对照组(P<0.01),第14天归芪组CD54表达水平下降,明显低于对照组(P<0.01)。照射后第8天归芪组CFU S计数显著高于对照组(P<0.01)。结论归芪口服液能够促进放射损伤小鼠早期骨髓造血功能的恢复,其机制之一可能是通过调节骨髓组织中黏附分子CD54的表达。 Objective To explore the effect of Gui Qi oral liquor on the expression of CD54 in bone marrow and its possible mechanisms of hematopoietic recovery in acute irradiation injured mice. Methods The acute irradiation injured mice were divided into 2 groups: Gui Qi group and control group, and 6 normal mice (normal group) received no treatment. After irradiation by 6.0 Gy ^60Coγ-ray, each mouse was administered with 200mg Gui Qi oral liquor through a gastric tube twice a day for 14 days in Gui Qi group, and equal quantity saline was given to each mouse in control group by the same way. On the 4th, 8th, 14th day after irradiation, the bone marrow mononuclear cells (BMNC), megakaryocytes in bone marrow tissue of mice were counted, and the proportion of hematopoietic tissue and the expression level of CD54 in bone marrow were evaluated. The colony forming units of spleen (CFU-S) of mice were counted on the 8th day after irradiation. Results On the 4th, 8th, 14th day after irradiation, the BMNC, megakaryocyte counts and the proportion of hematopoietic tissue in Gui Qi group were higher than those in control group (P〈0.01 or P〈0.05) . The CD54 expression level in Gui Qi group was higher than that in control group on the 4th, 8thday (P〈0.01), but lower than that in control group on the 14th day (P〈0.01) . On the 8th day, CFU-S counts in Gui Qi group were more than those in control group (P〈0.01) . Conclusion Gui Qi oral liquor can regulate CD54 expression in bone marrow of acute irradiation injured mice, which may be one of the mechanisms of Gui Qi oral liquor accelerating the early phase hematopoietic recovery of mice.
