摘要
目的研究白细胞介素8(IL8)在食管癌组织中的表达变化,以及此变化对肿瘤血管形成、患者预后的影响。方法利用实时逆转录聚合酶链反应(realtimeRT PCR)和免疫组织化学技术检测食管癌及正常组织中IL8mRNA和蛋白表达,并采用Ⅷ因子相关抗原(FⅧRAg)检测微血管密度。结果食管癌组织中IL8mRNA及蛋白表达显著高于正常组织。IL8mRNA高表达与远处淋巴结转移、微血管高密度、生存时间短(<26个月)、早期复发(<16个月)有关(均P<0.05)。癌组织中微血管密度比周围正常组织高(P<0.01)。结论在食管癌中,IL8mRNA高表达的患者发生转移机率高,复发早,生存时间短;IL8可以作为一个判定食管癌预后的指标。
Objective To study the expression of IL-8 in esophagus carcinoma and its relation with angiogenesis and prognosis. Methods By using the real time quantitative reverse-transcription-polymerase chain reaction and immunohistochemical method, the expression levels of IL-8 mRNA and protein in esophagus carcinoma and normal tissue were detected. By using Fac Ⅷ antigen, the microvessel density (MD) was measured. Results The expression levels of IL-8 mRNA and protein in esophagus carcinoma were significantly higher than in normal tissue. The high expression of IL-8 mRNA was correlated with distant lymph node metastasis, high tumor MD, short survival (〈26 moonths), and early relapse (〈16 months) (all P〈0.05) . MD in tumor tissue was significantly higher than in normal tissue (P〈0.001) . Conclusion In esophagus carcinoma, IL-8 mRNA expression is strongly associated with tumor angiogenesis, survival and relapse, suggesting it can be used as a prognostic indicator.
