三氧化二砷诱导人髓样甲状腺癌TT细胞凋亡

摘要 目的观察三氧化二砷(As2O3)对人髓样甲状腺癌TT细胞体外生长的影响。方法选用不同剂量As2O3处理人髓样甲状腺癌TT细胞后,通过倒置相差显微镜、电子显微镜、MTT法、AO/EB荧光染色法和流式细胞术研究As2O3对人髓样甲状腺癌TT细胞生长的影响。结果As2O3具有抑制人髓样甲状腺癌TT细胞生长和促进其凋亡的作用,这种作用主要在中浓度(2.0~5.0μmol/L)产生,而在高浓度(10.0μmol/L)则可引起细胞死亡。结论As2O3明显抑制人髓样甲状腺癌TT细胞生长,并诱导细胞凋亡,在甲状腺癌的辅助治疗上具有很好的应用前景。 To investigate the effect of arsenic trioxide (As2O3 ) on the growth of human myeloid thyroid cancer TT cells. Methods After treatment of TT cells with different concentrations of As2O3 in vitro, cell growth was assayed by inverted phase contrast microscope, scanning electron microscope, MTT method, AO/EB of fluorescent light staining and flow cytometry. Results As2O3 had an effect of inhibiting the proliferation and inducing apoptosis of TT cells. In middle concentration (2.0-5.0μmol/L), there mostly appeared the induction of apoptosis, while in high concentration (10.0μmol/L) there mostly appeared the cell death. Conclusion As2O3 can significantly inhibit the proliferation of TT cells and induce apoptosis of these cells. There is an excellent prospect for adjuvant therapy of thyroid cancer.
