摘要 目的研究白细胞介素5(IL5)对人嗜酸性粒细胞(Eos)中转化生长因子β1(TGFβ1)表达的调控作用。方法采用改良的密度梯度离心法分离并体外培养人外周血Eos,实验组加入相同浓度的白细胞介素4(IL4)、IL5和干扰素γ(IFNγ)以及不同浓度的IL5共同培养,ELISA和RT PCR法检测细胞培养上清液TGFβ1蛋白和mRNA的表达。结果与对照组相比,浓度均为10-9mol/L的IL4、IL5在蛋白水平和转录水平对TGFβ1的表达均有上调作用,而IFNγ则表现为抑制;10-11、10-9、10-7mol/LIL5均能显著增强体外培养的Eos中TGFβ1蛋白的表达。结论Th2型细胞因子IL5可以上调人嗜酸性粒细胞中TGFβ1的表达。 Objective To investigate the effects of IL-5 on the expression of TGF-β1 in human eosinophils in vitro. Methods Eosinophils were incubated in vitro in the presence of same concentrations of IL-4, IL-5 and IFNγ, or different concentrations of IL-5. After cultivation for 16h, the expression of TGF-β1 mRNA and protein in the supernatants was detected by ELISA and RT-PCR. Results As compared with the control, the expression of TGF-β1 protein and mRNA in eosinophils was increased significantly by IL-4 and IL-5, while decreased by IFNγ. At the sametime, the results demonstrated that the basal level of TGF-β1 expression in the cultured eosinophils in vitro was enhanced by IL-5 in all concentrations (10^11-10^-7mol/L). Conclusion IL-5 can up-regulate the expression of TGF-β1 in eosinophils in vitro.
