摘要
用绿色荧光蛋白(GFP)基因做报告基因,制备壳聚糖GFP质粒纳米粒,体外转染人肝癌细胞HepG2和肺癌细胞A549,定性评价纳米粒的转染效率和细胞毒性。结果表明壳聚糖GFP质粒纳米粒能转染两种细胞并在表达GFP,但两种细胞的转染效率不同,且该纳米粒能抑制两种细胞生长。
Chitosan-plasmid nanoparticles are prepared with GFP gene as reporter gene. and toxicity of the nanoparticles are investigated by transfecting HepG2 and A549 . The plasmid can be ferried into HepG2 and A549 and be translated into protein successfully. differently between HepG2 and A549. And the nanoparticles inhibit the proliferation of The transfection efficiency results show that the GFP But the cell uptake ratio is the two kinds cell lines.
