摘要
从黑曲霉固态发酵物中提取纤维素酶。粗酶液经硫酸铵盐析除去大量杂蛋白,然后通过2次SephadexG-100柱纯化CMC酶,提纯倍数可达47.22。酶学研究表明,纤维素酶中CMC酶最适作用温度为60℃,最适作用pH为4.5,CMC酶的Km为6.54mg/mL,Vmax为0.12mg/mL.min。
The cellulase is extracted from solid state leavening of Aspergillus niger LH2446. The crude product is purified by (NH4)2SO4 and chromatographed by Sephadex G-100 column gel. The CMC component of cellulase is 47.22 times of raw products. The study on the enzymology of CMC enzyme shows that the optimum reaction temperature is 60℃ , and pH is 4.5. The value of Km is 6.54 mg/mL and Vmax is 0.12 mg/mL/min.
