产几丁质酶木霉生防菌株的生化测定 被引量:21

Determination of biocontrol strain THS-1 of Trichoderma harzianum Producing Chitinase by biochemical methods

摘要 以烟草赤星病的病原真菌链格孢(Alternariaalternata)为实验菌株,通过对峙平板拮抗实验和胶体几丁质平板透明圈实验测定木霉菌株(Trichodermaharziaum)THS-1对病原真菌的拮抗作用,结合DNS法进行定量检测木霉菌株发酵产生几丁质酶的活性。上述方法的结合可以筛选出产几丁质酶高效木霉生防菌株。 THS-1, an isolate of Trichoderma harziaum, was proved to be strongly supprssive to tobacco brown spot pathogen Alternaria alternata (an isolate 97Aa003) by the dural culture method, and to be chitinase-producing by the clear zone method on colloidal chitin-containing agar plate. Also, chitinase produced by THS-1 was determined quantitatively by DNS(3,5-dinitrosalicylic acid) method. Thus, the biocontrol strains of Trichoderma producing chitinase were screened by the combination of the methods above.
