波尔山羊胚胎徒手分割试验

Trials of embryo splitting in Boer goat

在体视显微镜下,用含20%NCS的PBS液作为操作液,用显微玻璃针徒手分割波尔山羊早期囊胚。将13枚A级囊胚用于分割,其中11枚对称分割为两半胚,2枚为不对称分割。将13对半胚手术移植于13只同期发情的受体羊黄体侧子宫角,有5只受体妊娠,半胚移植受体妊娠率38.5%(5/13)。其中有2只妊娠受体发生流产,观察到3只流产胎儿,其余3只受体妊娠到期,获得3只正常半胚后代。

Free-splitting of early blastocysts were conducted under farm conditions in Boer goat.Embryos with good quality were placed in small droplet of phosphate buffered saline（PBS） containing 20 % new-born calf serurn（NCS）. Under a stereomicroscope, embryos with zona pellucidae were splitted on a vertical plane into two halves with a fine glass needle by hand. A total of 13 embryos were splitted, 11 double of them were symmetrical derni-embryos, and the other 2 double were not symmetrical demi-embryos. Demi-embryos with zonae were transferred surgically into 13 recipients uterine horn ipsilateral to the corpus luteum, 5 of them were pregnant, the pregnancy rate of demi-embryos transfer was 38.5 % （5/13）. Two of pregnant recipients were abort, and there were 3 abortus. Three pregnancy recipients went to term,and three demi-embryos kids were born.
