葛根素雌激素样作用的实验研究

Experimental study on the estrogen-like effect of puerarin

目的探讨葛根素的雌激素样作用。方法采用成年雌性SD大鼠,摘除双侧卵巢,分成6组假手术组(SHAM)、去卵巢组(OVX)、17β-雌二醇替代治疗组(ERT,0.5mg.kg-1.2d-1)、高剂量葛根素组(PH,100mg.kg-1.d-1)、中剂量葛根素组(PM,50mg.kg-1.d-1)和低剂量葛根素组(PL,20mg.kg-1.d-1)。去卵巢术后1周开始皮下注射给药,连续7周。通过观察大鼠阴道涂片情况、动情周期的改变、子宫内膜的变化及子宫系数的测定来观察葛根素的雌激素样作用。结果与OVX组相比,高、中剂量葛根素能使成年去卵巢大鼠子宫增重、阴道上皮细胞发生角化并增加动情周期的发生次数,但程度明显低于SHAM组及ERT组;而低剂量葛根素组无此改变。结论葛根素具有雌激素样作用,但作用强度弱于17β-雌二醇。

Objective： To evaluate the potential estrogen- like effect of puerarin. Methods： The adult female rats ovarieetomized （OVX） were divided into six groups： sham- operated group （SHAM）, OVX group, 1713 -estradiol replacement therapy group （ERT, 0. 5 mg·kg^-1·2 d^-1）, high- dose puerarin group （PH, 100 mg · kg^-1·d^-1）, middle- dose puerarin group （PM, 50 mg · kg^-1·d^-1） and low -dose puerarin group （PL, 20mg · kg^-1·d^-1） . The rats were given drugs by subcutaneous injection 1 week after OVX, lasting for 7 weeks, to observe vaginal smear, change of estrous cycle, changes of endometrium and detect uterine coefficient. Results： 100 and 50 mg · kg^-1·d^-1 injection of puerarin increased the weight of uterus, horuified vaginal epithelial cells and improved estrous cycle compared to those of OVX group, hut these changes were all lower than that of SHAM and ERT groups, and these changes were not seen in PL group. Conclusion： Puerarin has estrogen- like effect, but which is weaker than that of 17β -estradiol.
