采用猪肺巨噬细胞和Marc-145细胞培养对国内暴发流行PRRS的某地猪场进行了病毒分离,从流产胎儿分离出特征性致细胞病变因子。用PRRSV SDOW-17单克隆抗体进行间接免疫荧光染色,证明从4头流产胎儿分离出4株PRRSV(CH-la,CH-lb,CH-lc,CH-ld)。这4株毒均可在猪肺巨噬细胞和Marc-145细胞上生长,并产生特征性CPE变化。用PRRSV美国分离毒株VR-2332和NVSL抗血清分别对4株CH-1、VR-2332和NVSL毒株感染细胞进行间接免疫荧光试验,结果表明4个CH-l毒株近似于美洲型PRRSV。间接免疫荧光试验证明,4个CH-l分离株与HCV、PrV、PPV、TGEV、PEDV和HEV无交叉抗原。本文首次报道在国内暴发生殖和呼吸道综合征猪群分离到PRRS病毒。
Porcine alveolar macrophages and Marc-145 cells were used to isolate-the porcine repro-ductory and respiratory syndrome(PRRS)virus in pig herds with PRRS outbreaks. Four cy-topathogenic agents were isolated from 4 aborted fetuses, and were identified with PRRS monoclonal antibody SDOW-17 indirect fluorescent antibody(IFA) staining as PRRSV. The 4 isolates ,designated as CH-la , CH-lb,CH-lc , CH-ld .could replicate in both procine alveolar macrophages and Marc-145 cells,and cause cytopathognic effects(CPE)on the infected cells, Preliminary results of IFA staining with antisera to American strains of VR-2332 and NVSL indicated that the 4 isolates were antigenically similar to the American type of PRRS viruses. The 4 isolates did not react with HCV,PrV,PPV,TGEV,PEDV nd HEV antisera in the IFA tests. This is the first report on the isolation of PRRSV in China.
