胰腺癌组织中人端粒酶逆转录酶与P-糖蛋白和bcl-2蛋白关系的研究 被引量:3

摘要 目的:研究人端粒酶逆转录酶(human telom erase reverse transcriptase,hTERT)与P糖蛋白与bcl2蛋白在胰腺癌组织中的表达及其关系和临床意义。方法:用链霉抗生物素蛋白生物素过氧化物酶复合物法检测hTERT,链霉抗生物素蛋白过氧化物酶法检测P糖蛋白,链霉抗生物素蛋白生物素标记法检测bcl2蛋白,并用Pearson关联系数(rp)研究彼此的关联性。结果:在胰腺癌组织中hTERT的阳性率为82%,而在正常胰腺组织和慢性胰腺炎组织中无表达(均为P<0.001);P糖蛋白在胰腺癌组织中表达的阳性率为53%,明显高于慢性胰腺炎组织、正常胰腺组织的22%、17%(均为P<0.05);bcl2蛋白在胰腺癌组织中表达的阳性率为58%,而正常胰腺组织和慢性胰腺炎组织均无表达(均为P<0.001)。在胰腺癌组织中,hTERT的表达分别与P糖蛋白和bcl2蛋白的表达有关联性(P<0.05,P<0.001);P糖蛋白和bcl2蛋白的表达也有关联性(P<0.001)。结论:hTERT、P糖蛋白与bcl2蛋白在胰腺癌组织中均有较高的表达,hTERT与P糖蛋白和bcl2蛋白的表达分别具有关联性,提示hTERT在胰腺癌细胞中具有耐药和抗凋亡的作用,检测hTERT和bcl2蛋白有助于鉴别胰腺癌和慢性胰腺炎,而且可通过检测hTERT来预测化学治疗的效果。 Objective: To investigate the expressions of human telomerase reverse transcriptase (hTERT), P-glycoprotein (P-gp) and bcl-2 protein and their correlations with each other in pancreatic cancer tissues, and their clinical significances. Methods: Expressions of hTERT, P-gp and bcl-2 protein were tested by streptavidin-biotin-peroxidase complex, streptavidin- peroxidase and labelled streptavidin-biotin respectively. Their correlations were described by Pearson coefficient (rp). Results: The positive rate of expression of hTERT was 82% in pancreatic cancer tissues, but none in chronic pancreatitis tissues and normal pancreatic tissues (P 〈0. 001 ). The Positive rates of expression of P-gp were 53% , 22% and 17% in pancreatic cancer tissues, chronic pancreatitis tissues and normal pancreatic tissues respectively. The positive rate of expression of bcl-2 protein was 58% only in pancreatic cancer tissues. There were positive correlations between expression of hTERT and P-gp (P 〈0. 05 ) or bcl-2 protein (P 〈0. 001 ), and between expression of bcl-2 protein and P-gp(P 〈0. 001 ). Conclusion: There are higher expressions of hTERT, P-gp and bcl-2 protein in pancreatic cancer tissues than in others. There are positive correlations between expressions of hTERT and P-gp or bcl-2 protein. The results indicate that hTERT is of effects of resistant chemical-therapy drugs and anti-apoptosis in the human pancreatic cancer cells. Expression of hTERT and bcl-2 protein can distinguish pancreatic cancer from chronic pancreatitis and expression of hTERT can predict the effect of chemical therapy.
