摘要
目的观察刺梨提取物CL的体外抗肿瘤作用。方法采用溴化3-(4,5-二甲基噻唑2)-2,5-二苯基四氮唑蓝(3-[4,5-dimethylthiazol-2-yl]-2.5-diphenyltetrazolium bromide,MTT)检测法测定CL对胃癌-7901和MKN-45细胞的细胞毒性作用,台盼蓝排染法计数不同浓度药物作用下,不同时间点的活细胞数,绘制细胞生长曲线。结果MTT法检测显示,0.01μl·ml1~100μg·ml-1的CL 处理12~96h胃癌SGC-7901细胞和MKN-45细胞生长的抑制率均呈现剂量依赖性和时间依赖性, 细胞生长曲线显示:随CL浓度的升高曲线逐渐右移,且100μg·ml1的CL对SGC-7901细胞的生长曲线呈逐渐下降趋势。结论CL对胃癌7901和MKN-45细胞的体外生长具有一定的抑制作用,这种抑制作用均呈刺量依赖性和时间依赖性,提示CL有一定的体外抗肿瘤作用。
Objective To investigate the anticancer effect of CL of the extract of Rosa roxburghii Tratt on gastric carcinoma MKN-45 and SGC-7901 cells in vitro. Methods Cytotoxicity of CL on SGC-7901 and MKN-45 cells was measured by MTT reduction test. The number of living cells was counted by trypan blue dye exclusion method and growth curves were drawn according to it. Results The inhibitory percentages of CL (0.01μg·ml^-1-100μg·ml^-1) on growth of both SGC-7901 and MKN-45 cells were concentration-dependent and time-dependent from 12h to 96h. Growth curves show rightshift with concentration increasing. Especially, the growth curve of SGC-901 treated with CL at 100μg·ml^-1 was fall-ff. Condttsion CL has anticancer effect on SGC-7901 and MKN-5 cells in vitro.
