旱生香茶菜总二萜对荷人肝癌裸鼠皮下移植瘤生长的影响及其毒性研究

The effect and toxicity of Diterpenoids of Isodon Xerophilus(IXD) on the growth of human hepatocellular carcinoma cell line (CA) xenograft tumor in nude mice

目的研究IXD对荷人肝癌裸鼠皮下移植瘤生长的影响及其毒性。方法对荷人肝癌的裸鼠经灌胃给以IXD,观察其体内对人肝癌移植瘤生长的影响及对裸鼠主要脏器的毒性。结果对裸鼠人肝癌移植瘤增殖的抑制率和肝脏重量的影响,与阴性对照组比较IXD中、高剂量组均有显著性差异(P<0.05)。对裸鼠体重增加的抑制率IXD中、高剂量组与阳性对照组比较有显著性差异(P<0.01)。对脾、肾重量的影响,IXD低、中、高剂量组与阴性对照组比较均无显著性差异(P>0.05)。结论IXD对裸鼠人肝癌移植瘤生长有一定的抑制作用,IXD对裸鼠具有一定的毒性作用。

Objective To study the effect and toxicity of Isodon Xerophilus（IXD） on the growth of human hepatocellular carcinoma cell line （CA） xenograft tumor in nude mice. Methods Then nude mice were treated with IXD p.o for 12 days. The effects of IXD on the growth of human hepatocellular carcinoma, the weight and pathological changes of nude mice＇s main organs were observed. Results The inhibitory rate of IXD to the growth of human hepatocerlular carcinoma（CA） and the weight of liver, compared to the negative control, the above mentioned effects of IXD（100、200mg/kg） were of statistic significance（P〈0.05）. The inhibitory rate of IXD to the mouse weight, compared to the positive control（5-Fu）, the inhibitory effects of IXD（100.200mg/kg） were of statistic significance （P〈0.01）.Compared to the negative control, there were no significant difference to the weight of kidney and spleen at the doses of 50、100、200mg/kg.d^-1×12d via p.o. separately. Conclusions It could effectively inhibit the growth of nude mice xenograft hepatocellular carcinoma, while having some toxicity o^ally at certain dose rangs.
