摘要
目的:探讨人肺巨细胞癌高(95D)低(95C)转移株中细胞周期调控因子的表达水平与细胞周期/细胞增殖的关系。方法:MTT法检测两株细胞的增殖能力,流式细胞仪检测两株细胞的细胞周期差异,Western印迹检测两株细胞的周期调控因子的表达情况。结果:95D的细胞增殖能力显著高于95C细胞,与此一致,处于S期的95D细胞数目明显高于95C细胞;在蛋白质水平,95D细胞p27的表达低于95C细胞,而CDK2,磷酸化Rb的表达不同程度的高于95C细胞。结论:p27表达的不同,以及下游CDK2,Rb及Rb磷酸化水平的差异,最终引起两株细胞在细胞周期和细胞增殖能力的差异。
Purpose:To study the molecular mechanism that determines the cell cycle differences between human giant-cell carcinoma cell strains 95C and 95D.Methods:FACscan was used to analyze the cell cycle of 95C and 95D cells.and the proliferation ability of 95C and 95D cells was measured by MTT assay.We further detected the expression levels of cell eyele factors by Westem blotting.Results:Our data showed that the proliferation ability of 95D cells is higher than that of 95C cells,and that the cell number in S phase of 95D cells is much larger than that of 95C cells.We further found that the expression level of p27 in 95D cell is lower than that of 95C cells.In addition,the expression levels of CDK2 and p-RB of 95D cells are higher than that of 95C cells.Conclusions:Our resullts indieated that low expression level of p27 lead to the increased proliferation ability of 95D cells.Which might reflect the progreasion of human long cancer cells in cell eycle farets.
