摘要
Fenton反应体系中产生的羟自由基(·OH)攻击苯甲酸后,形成羟基苯甲酸,后者产生荧光并可定量检出。本文对该法进行了系统的条件试验,间羟基苯甲酸的直线线性范围0.0~9.0mg/L,CV<5%,验证了其灵敏度、可靠性和精密度,使其操作技术更符合简便化和规范化要求。首次提出一项可用于评价血清对·OH代谢的。影响能力的新指标—容受单位(X),建立了计算该指标的对数回归方程;(抑制率)=-32.13+52.73 lgX。30份血清样品的初步测定结果表明,正常人血清对·OH代谢的影响能力为9.7~20.7容受单位(95%可信区间)。
Benzoic acid is attracted by hydroxyl free radical (~·OH) in Fenton reaction to form hydroxybenzoic acid which can be quantitatively detected by means of fluorescence spectroscopic method. The sensitivity, reliability and precision (the linear limit for 3-hydroxybenzoic acid is 0.0~9.0mg/L, CV<5%) of the method were tested in order to make it standard and easy to be operated. Furthermore, a new parameter, capacity unit, for evaluating the effect of serum on the metabolism of ~·OH radical was advanced for the first time, and then a logarithmic regression equation, (reduced rate)=-32.13+52.73lg X (capacity unit), was esstablished. The results from the detection of 30 samples showed that the effect of the normal human serum on the metabolism of ~·OH radical was 9.7~20.7 capacity units (95%confidence limit).
