摘要
以中国白梨9个品种为实验材料,在分子水平上对其基因组DNA进行了PCR-RFLP系统检测、DNA序列测定及生物信息学分析,鉴定了6个品种的S基因型和3个品种的一个S等位基因,并从天生伏和蜜梨中分离鉴定了一个新的S等位基因,命名为梨S29-RN ase基因,该基因与S13的DNA序列最接近,推导氨基酸序列与S13仅有2个氨基酸序列的差异.
Pear tree is a typical GSI plant, and the SI is controlled by multiple allele gene in single S gene locus; so determination of S genotype is the basis of S1 study. In this paper, 9 cultivars of Chinese pear (Pyrus bretschnider) were studied by the method of PCR-RFLP and DNA sequence analysis. The genotypes of 6 cuhivars and one S allele gene of 3 cuhlvars were determined. Furthermore, a new gene, named S29. is isolated and determined from Tianshengfu and Mill. The new gene has the highest similarity to S13, and only two amino acids are different in their putative amino acid sequence.
