自交新高梨叶片的培养繁殖技术 被引量:13

摘要 以新高梨人工自交授粉所获种子经胚培养产生的试管苗为试材,采用正交试验设计,研究了基本培养基、TDZ、IAA和6-BA四因素及其组合对该自交种叶片不定芽再生的影响.结果表明,基本培养基的种类在愈伤组织分化出芽的过程中起着主导作用,而6-BA和IAA分别在诱导愈伤组织和诱导丛生芽阶段起主要作用.通过综合平衡法得出最佳培养基配方为NN69+0.5 mg*L-1 TDZ+0.3 mg*L-1 IAA;最佳接种试材为出芽后无菌培养15~30 d的叶片;最佳接种方式为叶基部向上插入接种. The test-tube plantlets of Niitaka pear attained from cultivating self-breed embryo by artificial pollination were selected as material. Usingorthogonal design, we researched the influence of the basic media. TDZ. IAA. and 6-BA on the budding frequency of the pure breed leaves. As a result, the species of the basic media is very important for budding form calli, and 6-BA. IAA are important for inducing calli and fasciculate buds respectively. Through synthesis and balance, we attained the best recipe of the culture medium, NN69+0. 5mg·L^-1 TDZ+0.3 mg·L^-1IAA; the best inoculating materials are leaves of 15-30 days' aseptic cultivation after budding and the best inoculating mode is to insert the medium with the leaves upwards from the base.
