摘要
目的:了解锌对人前列腺癌细胞增殖的影响。方法:采用不同浓度锌溶液干预人前列腺癌细胞株体外培养,测定细胞生长曲线,检测细胞生长抑制情况、细胞存活率、细胞凋亡及细胞周期情况。结果:锌对雄激素非依赖型(DU-145和PC-3)及雄激素依赖型(LNCaP)前列腺癌细胞均有抑制增殖和促进凋亡作用,对激素依赖型抑制更明显,3种细胞的IC50分别为780ng/ml、810ng/ml、100ng/ml。1000ng/ml锌对3种细胞生长均达最大抑制作用。随着锌浓度增高,3种细胞凋亡率均上升。结论:锌可以明显抑制雄激素非依赖型以及雄激素依赖型前列腺癌细胞的增殖,可能与锌诱导细胞凋亡有关。
Objective: To evaluate the effect of zinc on human prostatic carcinoma(PCa) cell growth. Methods: Growth inhibition and various regulatory responses were investigated in three human prostatic carcinoma cell lines (DU-145, PC-3 and LNCaP), treated with various concentrations of zinc. Results: Incubation of the PCa cell lines with different levels of zinc resulted in inhibition of cell growth, a lower 50 % inhibition of cell growth(ICS0) value for zinc(about 100 ng/ml)was detected in LNCaP cells, which were androgen-dependent, whereas androgen-independent PC-3 and DU-145 cells exhibited a higher IC50 for zinc(about 780 ng/ml and 810 ng/ml). Incubation with 1 000 ng/ ml zinc resulted in maximum inhibition of growth in both cell lines. Cell flow cytometric analyses revealed that the cell growth inhibitions were accompanied by an increase in apoptosis. Conclusion:These results suggest that zinc inhibits human prostatic carcinoma cell growth, possibly due to induction of cell apoptosis.
