喉癌及喉咽癌血管内皮生长因子表达 被引量:5

Expression of VEGF and VEGF-C in laryngeal and hypopharyngeal cancer

摘要 目的探讨喉癌、喉咽癌肿瘤组织中血管内皮生长因子家族成员VEGF(vascularendothialepithelialcellgrowthfactor)、VEGF-C的表达与肿瘤转移的相关性。方法患者46例,男35例,女11例,喉癌35例,喉咽癌11例,按照UICC1997年的TNM临床分类分期标准,Ⅰ期2例,Ⅱ期10例,Ⅲ期14例,Ⅳ期20例,T分级:T14例,T214例,T313例,T415例,N分级:N029例,N14例,N211例,N32例。所有患者均为M0级。既往未经任何治疗,病理均为鳞状细胞癌。抗VEGF、抗VEGF-C多克隆抗体SABC法免疫组化染色。结果VEGF在喉癌中的阳性表达率为78.26%(36/46),阳性表达与肿瘤的淋巴结转移及骨髓的微转移有明显的相关性;VEGF-C在喉癌中的表达率为73.91%(34/46),表达与肿瘤的淋巴结转移及骨髓微转移有相关性。结论血管生长因子家族的成员VEGF、VEGF-C参与肿瘤的血管及淋巴管的生长,促进肿瘤的转移。 OBJECTIVE To investigate the relationship of the expression of vascular endothial epithelial cell growth factor (VEGF, VEGF-C) on the metastasis of the laryngeal and hypopharyngeal cancer patients. METHODS We studied 46 laryngeal and hypopharyngeal cancer patients with no overt distant metastasis, who were treated in our hospital from December 2001 to February 2003. 35 of them were laryngeal cancer patients, 11 cases were with hypopharyngneal cancer. According to the UlCC 1987 TNM classification, there were 2 cases in stage Ⅰ, 10 in stage Ⅱ, 14 in stage Ⅲ, 20 in stage Ⅳ; T class: 4 with T1, 14 T2, 13 T3, 15 T4. N class: 29 with N0, 4 N1, 11N2, 2 N3. All patients were diagnosed as squamouse cell carcinoma. Anti-VEGF, anti-VEGF-C polyclone antibody and SABC immunohistochemistry method were employed to detect the expression in the tumor tissues. RESULTS The VEGF positive expression rate of the laryngeal and hypopharyngeal cancer was 78.26 %, and it had positive correlation with lymphoid and bone marrow metastasis. The expression of VEGF-C of the laryngeal and hypopharyngeal cancer tissue was 73.91%, and it correlated with the metastasis of the lymphoid nodes and bone marrow positively too. CONCLUSION The members of the vascular epithelial cell growth factors family VEGF and VEGF-C could promote the growth of the vessels in the tumor tissue, as a results it promotes the metastasis of the cancer.
