摘要
目的探讨乳腺浸润性导管癌中c-Myc、p27及细胞周期蛋白E(cyclin E)的表达、相互关系及其意义。方法应用免疫组化方法检测42例乳腺浸润性导管癌中c-Myc、p27及周期蛋白E蛋白的表达情况。结果c-Myc和周期蛋白E在42例乳腺浸润性导管癌中的阳性表达率分别为66·67%(28/42)和54·76%(23/42);p27的阴性率为52·38%(22/42)。28例c-Myc阳性表达的乳腺浸润性导管癌中周期蛋白E阳性表达的22例,p27阳性表达者8例;在20例p27阳性表达的乳腺浸润性导管癌中,周期蛋白E阳性6例,14例阴性。结论(1)c-Myc蛋白的表达与p27呈负相关关系,而与周期蛋白E的表达呈正相关;(2)p27的表达与周期蛋白E的蛋白表达间呈负相关;(3)乳腺浸润性导管癌中c-Myc对周期蛋白E/CDK2复合物活性的调控至少部分地依赖于c-Myc蛋白对p27蛋白的调节。
Objective To evaluate the significance of c-Myc, p27 and cyclin E expression in invasive ductal carcinoma of the breast and the relationship among the three proteins. Methods 42 cases with invasive ductal carcinoma of the breast were studied for c-Myc, p27 and cyclin E expression by immunohistochemical method. Results The positive rates of c-Myc and cyclin E in invasive ductal carcinoma of the breast were 66. 67% and 54.76% respectively,and the negative rate of p27 protein expression was 52.38%. Of 28 cases of invasive ductal carcinoma of the breast with c-Myc positive expression,22 cases had positive cyclin E expression and 8 cases p27 expression. Of 20 cases of invasive ductal carcinoma of the breast with p27 positive expression,6 cases were positive and 14 cases negative for cyclin E expression. Conclusions (1) There is a negative correlation between c Myc and p27,and a positive correlation between c-Myc and cyclin E. (2)A negative correlation between c-Myc and p27,and a positive correlation is found between p27 and cyclin E. (3)In invasive ductal carcinoma of the breast, the regulation of cyclin E/CDK2 by c-Myc is partly dependent on the regulation of p27 by c-Myc.
