摘要
以鄂柿1号甜柿离体新梢叶片为外植体,研究了秋水仙素浓度及处理时间对诱导染色体倍性变异的影响。结果表明,0.5%秋水仙素处理4d左右,可诱导获得十二倍体再生植株,低浓度的秋水仙素不能产生加倍效应,高浓度会导致叶片死亡或严重抑制愈伤组织再生能力;与对照比较,再生变异植株叶片气孔孔径增大、密度降低,PA倍性分析叶片DNA含量为对照的2倍。因此,试验获得的倍性变异株初步确认为12倍体。
A study of inducing polyploidy by treating in vitro leaves of Eshi 1 persimmon was carried out with different concentrations of colchicine solution and different treated durations. The results indicated that dodecaploid plantlets were obtained by treatment with 0.5% colchicine for about 4 days, and lower or higher concentration of colchicine was ineffective. Stoma characteristics and flow cytometry were used for identifying their ploidy levels which all showed differences between hexaploid and dodecaploid plantlets. The plantlets obtained in this experiment can be preliminarily affirmed to be dodecaploid.
