高效液相色谱法测定马桑叶中游离黄酮苷元 被引量:3

摘要 用反相高效液相色谱法,Shim-pack ODS柱,流动相为乙腈:水(冰乙酸调pH值为3.0)=35:65,v/v,流速1.0 ml/min,检测波长为360 nm,进样量15μl,以槲皮素为对照品,外标法定量,首次测定了马桑叶提取液中2种游离苷元-槲皮素、山奈酚在不同提取条件下的总含量.槲皮素进样量在0.0234~2.34 μg范围内与其峰面积呈良好线性关系,回归方程为Y(μg)=3.47355×10-7x+0.00393687,x2=0.999994.加标回收率为99.2%,RSD为1.4%. The free flavone aglycones in Coriaria sinica Maxim leaf were first determined by using reversed-phase high performance liquid chromatography, The HPLC system used ODS column with acetonitrile-water-acetic acid as the mobile phase at a flow rate of 1.0 ml/min and wave-length, detection of 360nm. The total contents of flavone aglycones-quercetin, kaempferol in the extracts of the leaves of Coriaria sinica Maxim under different conditions of extraction were determined by reference to external standard compound quercetin. A good linear relationship of standard was found in the range of 0. 0234 - 2.34μg with good correlation(r^2 = 0. 999994). Average recovery was 99.2 % (RSD = 1.4, n=6).
