重组ChIFN-γ增强鸡球虫细胞免疫作用的研究 被引量:18

摘要 初步探讨了重组鸡γ干扰素(rChIFN_γ)对鸡体抗球虫细胞免疫的增强效果,分别给7日龄和14日龄雏鸡邓E.tenella孢子化卵囊免疫并同时肌肉注射5 00 U rChIFN_γ后,于21日龄以同源E.tenella攻虫。试验结果显示:rChIFN_γ处理组的试验雏鸡脾脏和胸腺免疫器官指数显著高于免疫对照组(P<0.05),在21日龄时的淋巴细胞转化水平(1.14)也显著高于卵囊免疫对照组(0.95)(P<0.05);ABC染色结果表明rChIFN_γ可以显著增加试验雏鸡脾脏和盲肠的CD4+、CD8+T淋巴细胞数量;Griess(NO2-)结果表明:rChIFN_γ处理组在21日龄的NO2-含量(2.77 ug/mL)显著高于免疫对照组(2.14 ug/mL)(P<0.05);溶菌酶(LSZ)试验表明,21日龄时rChIFN_γ处理组的LSZ含量(22.80 ug/mL)显著高于免疫对照组(19.79 ug/mL)(P<0.05)。结果提示rChIFN_γ具有抗球虫免疫的增强效果,能显著提高鸡体抗球虫保护性细胞免疫水平。 The regulatory effects of recombinant chicken interferon gamme( rChIFN-γ)on the cellular immunity of chickens vaccinated with E. teneUa were studied in this paper. Chickens vaccinated with E. tenella spornlated oocysts at 7 and 14-day old respectively were simultaneously injected with 5 000 U rChIFN-γ, and were challenged with 5 × 10^4 homologous E. tenella sporilated oecysts at 21-day old respectively for detecting the parameters. The results showed that rChIFN-γ can increase the immune organ index of treated chickens. Lymphocyte prnliferatin assays(LPA)levels of chickens injected with rChIFN-γ(0.81) were significantly higher than the control chickens (0.67) immunized only with oocysts. The increasing of the numbers of CD^4+ , CD^8+ T cell in spleen and ceacum of chickens injected with rChIFN-γ were found through the ABC technique.Te results of Griess response showed that rChIFN)γ can significantly increase NO2^- level in chickens treated (P〈0.05).LSZ level of chickens immunized with rChIFN-γ (22.80 ug/mL) was also significantly higher than immunized control chickens vaccinated only by oecysts( 19.79 ug/ml).The results implied that rChIFN-γ is an important cytokinc which has the ability to enhance the protective immunity against the coecodiosis, and could he an excellent potential adjuvant for anticoccodial vaccination.
